21-Gene Recurrence Score Assay Could Not Predict Benefit of Post-mastectomy Radiotherapy in T1-2 N1mic ER-Positive HER2-Negative Breast Cancer

Conclusion: This study showed that the 21-gene RS assay was not able to predict the benefit of PMRT for OS in women with T1-2 N1mic ER-positive HER2-negative breast cancer. However, further prospective larger sample-size trials are warranted to determine if a benefit exists. Introduction According to the latest Global Cancer Statistics 2018, breast cancer is still the most frequent cancer and the leading cause of cancer death among females worldwide (1). It is estimated that there will be about 2.1 million newly diagnosed female breast cancer cases and 627,000 cancer death in 2018 (1). The use of screening tests for breast cancers provides better opportunities for patients to detect disease at an earlier stage and obtain more effective treatment with fewer side effects. As reported by the National Cancer Institute (NCI) in the Surveillance, Epidemiology, and End Results Program (SEER), about 93% of female breast cancers are diagnosed at localized or regional stage1. For early-stage breast cancer patients, three landmark clinical trials have demonstrated that post-mastectomy radiotherapy (PMRT) reduces locoregional recurrences (LRRs) and improves overall survival (OS) in high-risk patients, namely those with pathologically involved axillary nodes or with large tumor size (>5 cm) (2–4).
