(Cincinnati Children's Hospital Medical Center) Cell-by-cell genetic analyses of developing brain tissues in neonatal mice and laboratory models of brain cancer allowed scientists to discover a molecular driver of the highly aggressive, deadly, and treatment-resistant brain cancer, glioblastoma. Published in Cell Stem Cell, the findings present an opportunity to find out if new therapeutic approaches can stop glioblastoma at its earliest stages of initial formation or recurrence.

In this study, we confirm our initial findings from the proteomic screen and demonstrate that LASP1 can interact with both eIF4A and eIF4B. Importantly, the LASP1-eIF4A and LASP1-eIF4B interaction is shown to be CXCL12-dependent. In addition, the ability of CXCR4 to impact the phosphorylation of eIF4F regulatory proteins is provided. Taken together, we hypothesize that activation of CXCR4 can promote eIF4F complex formation and activity through LASP1 and cell signaling. In conclusion, we showed hypermethylation of CpGs as a novel mechanism of action for DNMTi agents and identified 638 hypermethylated molecular targets (CpGs) common to decitabine and azacytidine therapy.
