Qingpeng Ointment Ameliorates Inflammatory Responses and Dysregulation of Itch-Related Molecules for Its Antipruritic Effects in Experimental Allergic Contact Dermatitis

In this study we used HPLC and LC/MS analysis, combined with a BATMAN-TCM platform, for detailed HPLC fingerprint analysis and network pharmacology of QP, and investigated the anti-inflammatory and antipruritic activities of QP on ACD induced by squaric acid dibutylester (SADBE) in mice. The BATMAN-TCM analysis provided information of effector molecules of the main ingredients of QP, and possible chronic dermatitis-associated molecules and cell signaling pathways by QP. In ACD mice, QP treatment suppressed the scratching behavior induced by SADBE in a dose-dependent manner and inhibited the production of Th1/2 cytokines in serum and spleen. Also, QP treatment reversed the upregulation of mRNAs levels of itch-related genes in the skin (TRPV4, TSLP, GRP, and MrgprA3) and DRGs (TRPV1, TRPA1, GRP, and MrgprA3). Furthermore, QP suppressed the phosphorylation of Erk and p38 in the skin. In all, our work indicated that QP can significantly attenuate the pathological alterations of Th1/2 cytokines and itch-related mediators, and inhibit the phosphorylation of MAPKs to treat the chronic itch. Nusinersen can treat patients with spinal muscular atrophy by modulating alternate splicing of the SMN2 gene and increasing survival of motor neuron protein levels. Since the approval of nusinersen in Japan in 2017, we have evaluated such patients to understand the changes in the clinical course after completed treatment. Currently, the factors correlating with good and poor responses to nusinersen treatment remain unclear. We retrospectively assessed the clinical outcomes of our treated cohort, in relevance to the level of muscle damage semiquantified by muscle MRI performed before the treatment.

Satellite cells are residential muscle stem cells that express a paired box transcription factor PAX7. Pax7 has been shown to be involved in determination of myogenic cell lineage during development, yet it is also required for the postnatal muscle growth, muscle regeneration and maintenance of the satellite cell pool. Although the functions of Pax7 have been studied thoroughly, its interacting partners in myogenic cells in vivo are largely unknown due to the lack of proper antibodies for immunoprecipitation.

Urine-derived stem cells (USCs) are known to be a very promising source of stem cells, in addition their isolation is non-invasive, relatively quick and simple, affordable, ubiquitous, and readily accepted by patients. The USCs transcriptional profiling showed a great variability across individuals probably due to the co-presence of distinct cell types. The lack of specific and unique cell surface markers for the identification of cell types makes the separation difficult. In order to separate the USCs sub-populations, we used the new technology Celector that sorts the cells based only on their physical characteristics.
