AqeilanSummaryDNA double-strand breaks (DSBs) are deleterious and tumorigenic but could also be essential for DNA-based processes. Yet the landscape of physiological DSBs and their role and repair are still elusive. Here, we mapped DSBs at high resolution in cancer and non-tumorigenic cells and found a transcription-coupled repair mechanism at oncogenic super-enhancers. At these super-enhancers the transcription factor TEAD4, together with various transcription factor...

Publication date: 15 October 2019Source: Cell Reports, Volume 29, Issue 3Author(s): Sneha Saxena, Suruchi Dixit, Kumar Somyajit, Ganesh NagarajuSummaryATR kinase-mediated replication checkpoint is vital for genome maintenance following replication stress. Previously, we showed that XRCC2-RAD51D (DX2) sub-complex of RAD51 paralogs restrains active DNA synthesis during dNTP alterations, in a manner dependent on ATR-mediated phosphorylation of XRCC2. Here, we find that unrestrained fork progression in XRCC2 deficiency and phosphorylation defect causes replication-associated errors, subsequently resulting in genome-wide double...

Abstract BACKGROUND: Inadequate self-care is linked to poor health outcomes in heart failure (HF). Self-care depends on decision-making abilities, but links between self-care and brain injury to executive decision-making regulatory areas (prefrontal cortices) are unclear. OBJECTIVE: We investigated the relationships between HF self-care and status of prefrontal cortices. METHODS: Magnetic resonance imaging-based diffusion tensor imaging was performed in 21 patients with HF (age, 53.8 ± 7.9 years; 15 men; left ventricular ejection fraction, 25.1% ± 6.1%), and self-care and executive function...

A comprehensive analysis of the nucleotide selectivity by the mitochondrial RNA polymerase (RNAP) RpoTm, from Arabidopsis thaliana, has been carried out. The kinetic parameters for the incorporation of cognate, non-cognate and oxidized bases have been determined. The results establish high fidelity of mitochondrial transcription resembling those of replica...

This article centers on two families of sequence-specific transcription factors: bZIP (basic leucine-zipper) proteins, exemplified by the AP-1 and CEBPβ recognition of 5mC; and bHLH (basic helix-loop-helix) proteins, exemplified by MAX and TCF4 recognition of 5caC. We discuss the impact of template strand DNA modification on the activities of DNA and RNA polymerases, and the varied tendencies of modifications to alter base pairing and their interactions with DNA repair enzymes.Graphical abstract

Publication date: Available online 15 October 2019Source: Journal of Molecular BiologyAuthor(s): Ke Liu, Jinrong MinAbstractCXXC domain-containing proteins are often involved in different biological processes, such as cell proliferation and development, apoptosis, DNA repair, signaling transduction and tumourigenesis, by regulating transcription. 12 CXXC domain-containing proteins have been identified in human, and structural together with DNA binding studies reveal that these CXXC domains, a 50-70 residues module of two conserved CXXCXXC motifs, bind to non-methylated DNA in different sequence contexts. These CXXC domain-...

Syam PrasadAbstractAluminium doped phosphate based bioglasses have potential applications in the field of bone tissue engineering because of their excellent bioactivity and biocompatibility along with high mechanical strength and controlled dissolution. In the present study, 8ZnO–22Na2O–(24-x)CaO–46P2O5–xAl2O3 (where x = 0, 2, 4, 6, 8 and 10 mol%) glass system was synthesized and investigated by means of XRD, FTIR, SEM and...

CONCLUSIONS: The incidence of hemopericardium complicating transseptal access during BMV was 2.0%, and was associated with a mortality rate of 6.9%. BMV can be safely performed in the same sitting with a second transseptal puncture, in patients with a favorable valve morphology. Surgical intervention can be reserved for a subset of patients with persistent pericardial collection. PMID: 31611427 [PubMed - as supplied by publisher]
