AbstractPurposeThe role of cytokine-producing B cells in antitumor immunity is mostly overlooked. In the present study, we investigated changes in B cell cytokine profiles in breast tumor-draining lymph nodes (TDLNs) during disease progression, and associations of these changes with prognostic indicators.MethodsFlow cytometry was used to measure the expression of TNF- α, IL-10, TGF-β, IL-2 and IFN-γ in B cells from 42 axillary lymph nodes. ConclusionsDual-labeled trastuzumab showed specific accumulation in orthotopic HER2-positive BT474 breast tumors with micro-SPECT/CT and fluorescence imaging and enabled image-guided tumor resection. In the clinical setting, [111In]In-DTPA-trastuzumab-IRDye800CW could be valuable for preoperative detection of (metastatic) tumors by SPECT/CT imaging, and intraoperative localization by using a gamma probe and fluorescence image-guided surgery to improve radical resection of tumor tissue in patients with HER2-positive tumors.

This article intends to summarize personal highlights from the 2019 ASCO (American Society of Clinical Oncology) Annual meeting. The article does not aim to offer a  comprehensive summary of the 1642 abstracts presented, but rather aims to highlight the abstracts that are the most relevant for the neo/adjuvant therapy of the HER2-negative, HR-positive breast cancer. Amplification and overexpression of human epidermal growth factor receptor 2 (HER2) in breast cancer is associated with an adverse prognosis. The introduction of trastuzumab and lapatinib has substantially improved the clinical outcomes of patients with HER2-positive breast cancer. The key element of the successful application of anti-HER2 therapies in real-world has been the selection of candidates for treatment based on the level of HER2 positivity of the tumor.
