In a study reported by Zhu et  al (Circulation 2018;138:2682–2697, PMID 30571258), the investigators identified a rare, heterozygous ankyrin-B (ANK2) variant (p.Q1283H) in a proband with recurrent ventricular tachycardia. In order to determine the functional effects of the ANK2 variant, knockin (KI) mice harboring the p.Q1283 H variant were generated. The KI mice showed increased ventricular arrhythmias after catecholaminergic stimulation. Isoproterenol resulted in increased frequency of delayed afterdepolarizations and Ca2+ waves and sparks as well as decreased sarcoplasmic reticulum Ca2+ content in KI cardiomyocytes.
