Study Eases Concern That Common Diabetes Drug Canagliflozin (Invokana, Invokamet) Might Harm Bones

THURSDAY, Jan. 3, 2019 -- If you have type 2 diabetes and you're taking canagliflozin to help control your blood sugar, a new study has some good news for you: The drug doesn't appear to raise the risk of bone fractures. Previously, research had...

Obesity (Silver Spring). 2021 Sep 21. doi: 10.1002/oby.23249. Online ahead of print.ABSTRACTDysregulation of hepatic glucose production (HGP) and glucose disposal leads to hyperglycemia and type 2 diabetes. Hyperglycemia results from the declining ability of insulin to reduce HGP and increase glucose disposal, as well as inadequate ß-cell compensation for insulin resistance. Hyperglucagonemia resulting from reduced suppression of glucagon secretion by insulin contributes to hyperglycemia by stimulating HGP. The actions of pancreatic hormones are normally complemented by peptides secreted by cells distributed througho...

We report that preadipocyte differentiation increased the abundance of mRNA levels of proteins related to adipogenesis and lipid metabolism. These changes in the transcriptome were absent or less pronounced in fat cells obtained from patients with prediabetes and type 2 diabetes. Meanwhile, mRNA levels of proteins involved in excellular matrix remodeling, angiogenesis and cytoskeleton organization were less abundant in adipocytes of all metabolic groups.

In this study, we explored the association between miR-204 and β-cell dysfunction, the therapeutic effects of berberine (BBR) on β-cell function and the possible mechanisms. We found that miR-204 increased and SIRT1 mRNA and protein levels decreased significantly in islets both in vivo and in vitro. MIN6 cells induced by palmitic acid exhibited increased apoptosis, and the accumulation of insulin and ATP in the supernatant decreased. Importantly, palmitic acid treatment combined with miR-204 silencing showed opposite changes. MiR-204 overexpression in MIN6 cells increased apoptosis and decreased insulin and ATP p...

Men and women have different type 2 diabetes mellitus (T2DM) risks, which have been reported across populations of different ethnicity. Where differences in T2DM risk for sex (biological) have been studied, research on gender (socio-cultural) and T2DM risk is lacking. We explored, in a multi-ethnic population, the association of six gender-related characteristics with incident T2DM over 3 years, and the mediation by known risk factors for T2DM.

ConclusionsSupervised ML is an innovative tool for the development of new methods to exploit large health administrative databases. In context of InfAct project, we have developed and applied the first time a generic ML-algorithm to estimate the incidence of diabetes for public health surveillance. The ML-algorithm we have developed, has a moderate performance. The next step is to apply this algorithm on SNDS to estimate the incidence of type 2 diabetes cases. More research is needed to apply various MLTs to estimate the incidence of various health conditions.
