Emerging role of non-canonical polycomb repressive complexes in normal and malignant hematopoiesis

Polycomb group (PcG) proteins comprise the multiprotein complexes, polycomb repressive complexes (PRCs), which play an important role in the transcriptional repression of target genes through histone modifications. Two major complexes have been extensively characterized: PRC1 and PRC2, which add mono-ubiquitination at lysine 119 of histone H2A (H2AK119ub1) and mono-, di-, and tri-methylation at lysine 27 of histone H3 (H3K27me1/me2/me3), respectively [1] (Figure 1). PRC2 is recruited to non-methylated CpG islands (CGIs) through a not yet fully understood mechanism.
