Conclusions Because the expression of copper tolerance genes is critical for the survival and virulence of fungal pathogens, pharmacological disruption of the function of copper tolerance genes would be predicted to have the following beneficial effects: (1) Induce fungal death: the loss of copper tolerance genes would lead to toxic levels of copper that would be expected to have fungistatic and/or fungicidal effects on the organism. We report that the bifunctional compounds enhance the interactions between primary human neutrophils and A. fumigatus in vitro, using three microfluidic assay platforms. The bifunctional compounds significantly enhance the recruitment of neutrophils, increase hyphae killing by neutrophils in a uniform concentration of drug, and decrease hyphal tip growth velocity in the presence of neutrophils compared to the antifungal targeting moiety alone. Abstract In vitro pharmacokinetic/pharmacodynamic data of liposomal amphotericin B (L-AMB) were compared with animal data from neutropenic and non-neutropenic models of azole-susceptible and azole-resistant invasive aspergillosis. L-AMB was equally effective. The in vitro fCmax/MIC ratio associated with 50% of maximal activity was 0.31 (0.29-0.33) similar to neutropenic but not to non-neutropenic mice [0.11 (0.06-0.20)].
