Fibroblast Compression and Tumor Cells Migration

Role of Compression in MetastasisOurHuman Pancreatic Fibroblasts play a key role in this study.Pancreatic fibroblasts are continuously gaining ground as an important component of tumor microenvironment that dynamically interact with cancer cells to promote tumor progression. In addition, these tumor-infiltrated fibroblasts can acquire an activated phenotype and produce excessive amounts of extracellu lar matrix creating a highly dense stroma, a situation known as desmoplasia. Maria Kalli, Panagiotis Papageorgis, Vasiliki Gkretsi, Triantafyllos Stylianopoulos. (2018).Solid Stress Facilitates Fibroblasts Activation to Promote Pancreatic Cancer Cell Migration. Annals of Biomedical Engineering. 1. A schematic of the in vitro transmembrane pressure device. (a) Fibroblasts were grown as a monolayer on the transmembrane of a 0.4 lm transwell insert and a piston of adjustable weight was applying a compressive stress. Control cells were covered with an agarose cushion only. (b) The experimental set-up of the co-culture system consisted of fibroblasts and pancreatic cancer cells (MIA PaCa-2 or CFPAC-1) in the upper and lower chamber of a transwell insert, respectively. A piston with adjustable weight, applying 4.0 mmHg of compressive stress on fibroblasts for 48 h is shown. A co-culture system consisting of fibroblasts and cancer cells without a compressive load was used as a control.FIGURE 2. Neuromics '  Human Pancreatic Fibrob...

ConclusionsThe biological effects regulated by Silibinin can be mediated by JNK/SAPK signaling. This provides a solid theoretical basis for the role of Silibinin in the treatment of pancreatic cancer.

bino De Placido Roberto Bianco The optimal therapeutic strategy for locally advanced pancreatic cancer patients (LAPC) has not yet been established. Our aim is to evaluate how surgery after neoadjuvant treatment with either FOLFIRINOX (FFN) or Gemcitabine-NabPaclitaxel (GemNab) affects the clinical outcome in these patients. LAPC patients treated at our institution were retrospectively analysed to reach this goal. The group characteristics were similar: 35 patients were treated with the FOLFIRINOX regimen and 21 patients with Gemcitabine Nab-Paclitaxel. The number of patients undergoing surgery was 14 in the FFN grou...

AbstractSecreted frizzled ‐related protein 4 (SFRP4) is a member of secreted protein family with sequence similarity to frizzled receptors of wingless‐related integration site (Wnt) signaling pathways. These proteins control diverse functions from embryonic development to adults in many organisms including humans. Initia lly, SFRPs were recognized as antagonists of Wnt signaling and supposed to interact with Wnts. Further research demonstrated their interactions to frizzled receptors and a functional diversity was related to these proteins, Wnt signaling potentiation in addition to modulation. SFRP4 is the largest m em...

Objective: To define the influence of preoperative immune modulating nutrition (IMN) on postoperative outcomes in patients undergoing surgery for gastrointestinal cancer. Background: Although studies have shown that perioperative IMN may reduce postoperative infectious complications, many of these have included patients with benign and malignant disease, and the optimal timing of such an intervention is not clear. Methods: The Embase, Medline, and Cochrane databases were searched from 2000 to 2018, for prospective randomized controlled trials evaluating preoperative oral or enteral IMN in patients undergoing surger...

Objective: The aim of the study was to identify the survival of patients with locally advanced pancreatic cancer (LAPC) and assess the effect of surgical resection after neoadjuvant therapy on patient outcomes. Background: An increasing number of LAPC patients who respond favorably to neoadjuvant therapy undergo surgical resection. The impact of surgery on patient survival is largely unknown. Materials and Methods: All LAPC patients who presented to the institutional pancreatic multidisciplinary clinic (PMDC) from January 2013 to September 2017 were included in the study. Demographics and clinical data on neoadjuva...

Besselink the Dutch Pancreatic Cancer Group and International Collaborative Group on Locally Advanced Pancreatic Cancer Locally advanced pancreatic cancer (LAPC) has several definitions but essentially is a nonmetastasized pancreatic cancer, in which upfront resection is considered not beneficial due to extensive vascular involvement and consequent high chance of a nonradical resection. The introduction of FOLFIRINOX chemotherapy and gemcitabine-nab-paclitaxel (gem-nab) has had major implications for th...
