Bioactive aporphine alkaloids from Fissistigma tungfangense

Publication date: June 2018 Source:Phytochemistry Letters, Volume 25 Author(s): Qi Zhou, Yan-Hui Fu, Yu-Qin Zhang, Shou-Yuan Wu, Xiao-Ping Song, Wei Xu, Guang-Ying Chen Three new aporphine alkaloids, fissitungfines C–E (1–3), together with five known analogues (4–8) were isolated from the stems of Fissistigma tungfangense. Their structures were elucidated by extensive spectroscopic methods Among them, fissitungfines C (1) and D (2) are two unusual aporphine alkaloids with a carboxyl group as a substituent at N-6. All isolated compounds were evaluated for their anti-rheumatoid arthritis activities via examining their anti-proliferative effects on synoviocytes in vitro. Fissitungfine E (3) exhibited a significant inhibitory effect on the proliferation of synoviocytes comparable to that of methotrexate. Graphical abstract

Authors: Cui XF, Cui XG, Leng N Abstract Papillary renal cell carcinoma (PRCC) accounts for about 10 percent of all renal cell carcinomas, and the prognosis is poor for people with advanced disease. Interleukin-20 receptor subunit beta (IL20RB) is a single-pass type I membrane protein of the type II cytokine receptor family and is related to the pathogenesis of chronic inflammation and autoimmune diseases, including psoriasis, glaucoma, vitiligo, rheumatoid arthritis, and inflammatory bowel disease. However, little has been reported on IL20RB with respect to cancer, especially in PRCC. Thus, we performed this study...

Over the last two decades the improvement of knowledge of the pathogenetic mechanisms leading to the development of the disease has profoundly changed the treatment strategies of RA through the development of biotechnological drugs (bDMARDs) directed towards specific pro-inflammatory targets involved in...

This study evaluated spebrutinib pharmacology and mechanism of action over a 4-week treatment period in patients with active rheumatoid arthritis (RA).MethodsPrimary human B cells, T cells, natural killer cells, macrophages, dendritic cells, basophils, and osteoclasts were treated with spebrutinib in vitro. Clinical pharmacodynamics were studied in 47 patients with active RA on background methotrexate therapy randomized to oral spebrutinib 375  mg/day or placebo.ResultsIn vitro, spebrutinib inhibited B-cell proliferation more potently than T-cell proliferation and reduced both lymphoid and myeloid cytokine production ...
