Activation of the transforming growth factor-β (TGF-β)-mediated signaling pathway plays a key role in the scar pathogeneses, and high temperature requirement factor A1 (HTRA1) inhibits TGF-β1 activation in tumor cells. Our study aims to investigate the role of HTRA1 in the pathogenesis of scars. The mRNA levels of HTRA1 was evaluated by real time PCR, HTRA1 protein expression was determined using western blot and immunohistochemistry, and a luciferase assay was applied to measure dynamic changes of TGF-β1 activity. We found that the expression of HTRA1 was significantly elevated in keloid tissues, compared to normal skin, and TGF-β1 mRNA levels slightly increase in the keloid tissue. Furthermore, active TGF-β1 protein levels and Smad2 phosphorylation significantly increased in the keloid tissue. Treatment with the latent TGF-β1 or recombinant human HTRA1 (rhHTRA1), alone or in combination, increased Smad2 phosphorylation levels in keloid fibroblasts and active TGF-β1 contents of associated supernatants. To the Editor: We would like to comment on the recently published retrospective review by Renz et  al on adjuvant radiation therapy (RT) for excised keloids.1 Most patients in this series received 20 Gy in 5 fractions at 1 institution versus 12 Gy in 3 fractions or 16 Gy in 4 fractions at a second institution. Three other schedules were also used (18 Gy in 3 fractions, 15 Gy in 3 fractions , and 10 Gy in 2 fractions) for small numbers of patients. CONCLUSION Blistering at the postoperative site seems to be uncommon and particularly unlikely to occur in localized forms of EB. Postoperative wound infections and dehiscence are uncommon. Patients with EB may have a propensity to develop keloid or hypertrophic scarring. With these data, the authors hope clinicians have greater confidence in referring patients with EB for surgery, and surgeons more reassured about postoperative wound healing.

(Henry Ford Health System) Findings from a new study conducted by researchers at Henry Ford Health System suggest a link between keloids and increased risk of being diagnosed with breast cancer, particularly among African Americans. Keloids are benign fibroproliferative tumors, which can result in excessive growths of scar-like tissue on the skin. Keloids have been reported to affect some 11 million people worldwide. Despite this prevalence, the exact cause of keloid formation is unknown.
