Pyrroline-5-carboxylate reductase 1 promotes proliferation and inhibits apoptosis in non-small cell lung cancer.

Pyrroline-5-carboxylate reductase 1 promotes proliferation and inhibits apoptosis in non-small cell lung cancer. Oncol Lett. 2018 Jan;15(1):731-740 Authors: Cai F, Miao Y, Liu C, Wu T, Shen S, Su X, Shi Y Abstract Disordered tumor cell metabolism is involved in the process of tumorigenesis. Proline metabolism is of critical importance for tumor cells, and pyrroline-5-carboxylate reductase 1 (PYCR1), a key proline biosynthesis enzyme, has been reported to be overexpressed in prostate cancer and to promote tumor cell growth in breast cancer. The present study investigated the relationship between PYCR1 and non-small cell lung cancer (NSCLC). The results revealed that PYCR1 was overexpressed in NSCLC tumor tissues compared with adjacent normal tissues. High PYCR1 expression was associated with poor prognosis in patients with NSCLC. Following knockdown of PYCR1 by small interfering RNA, cell proliferation was revealed to be significantly inhibited and the cell cycle was arrested, while apoptosis was increased in SPC-A1 and H1703 NSCLC cells. Furthermore, the silencing of PYCR1 resulted in the downregulation of expression of the cell cycle regulator cyclin D1, the regulator of the mitochondrial apoptotic pathway B-cell lymphoma-2, and B-cell lymphoma-extra large. The results of the present study indicated the involvement of PYCR1 in the proliferation and apoptosis of NSCLC. Therefore, PYCR1 may be a novel therapeutic target for inhibiting cell proliferation in...

This report warns that reluctance to talk honestly about death is preventing people with cancer from dying'well'. It calls for Governments across the UK to honour their commitments to ensuring Advance Care Planning is used as an important part of a person-centred health service, so that people approaching the end of their lives receive the best care possible and their wishes for death and dying are fulfilled

The prognostic value of high sensitivity C-reactive protein in a multi-ethnic population after>10 years of follow-up: The Multi-Ethnic Study of Atherosclerosis (MESA). Int J Cardiol. 2018 Aug 01;264:158-164 Authors: Cainzos-Achirica M, Miedema MD, McEvoy JW, Cushman M, Dardari Z, Greenland P, Nasir K, Budoff MJ, Al-Mallah MH, Yeboah J, Blumenthal RS, Comin-Colet J, Blaha MJ Abstract BACKGROUND: The prognostic value of hsCRP in contemporary multi-ethnic populations is unclear, particularly in statin users. The aim of this study was to characterize the prognostic utility of hsCRP for atheroscleroti...

Abstract The molecular pathogenesis and classification of colorectal carcinoma are based on the traditional adenomaecarcinoma sequence, serrated polyp pathway, and microsatellite instability (MSI). The genetic basis for hereditary nonpolyposis colorectal cancer is the detection of mutations in the MLH1, MSH2, MSH6, PMS2, and EPCAM genes. Genetic testing for Lynch syndrome includes MSI testing, methylator phenotype testing, BRAF mutation testing, and molecular testing for germline mutations in MMR genes. Molecular makers with predictive and prognostic implications include quantitative multigene reverse transcriptas...

Abstract MicroRNAs (miRNAs) are poised to provide diagnostic, prognostic, and therapeutic targets for several diseases including malignancies for precision medicine applications. The miRNAs have immense potential in the clinical arena because they can be detected in the blood, serum, tissues (fresh and formalin-fixed paraffin-embedded), and fine-needle aspirate specimens. The most attractive feature of miRNA-based therapy is that a single miRNA could be useful for targeting multiple genes that are deregulated in cancers, which can be further investigated through systems biology and network analysis that may provid...

A 69-year-old man was referred to our department because of renal masses of the left kidney. The patient was diagnosed with a bladder carcinoma 10 months before in the context of macroscopic hematuria. The cancer had a partial squamous and papillary differentiation and was staged as G3pT1B with the infiltration of the superficial part of the bottom of the chorion. The patient underwent a transurethral resection of the bladder and had a course of intravesical injections of Bacillus Calmette-Guerin (BCG).

CONCLUSIONS: Our work supports the hypothesis that genetic variation of clock genes might affect cancer risk. These findings also highlight the need for more efforts in this research field in order to fully establish the contribution of clock gene variants to the risk of developing cancer. METHODS: We conducted a systematic review and meta-analysis of the evidence on the association between clock genes' germline variants and the risk of developing cancer. To assess result credibility, summary evidence was graded according to the Venice criteria and false positive report probability (FPRP) was calculated to further vali...

Conclusions: BIND-510 exhibits differentiated PK, tumor accumulation, tolerability and anti-tumor activity compared to VCR. In combination with the demonstration of PSMA expression in hematological cancers, our findings support the feasibility of developing BIND-510 as a targeted clinical therapy with the potential benefit of reduced toxicity and improved anti-tumor activity compared to currently available treatments in hematological as well as solid tumor settings.Citation Format: Louise Cadzow, Kristen Arnold, Daniel Thrasher, James Nolan, Allen Horhota, Eric Lewis-Clark, James Wright, Susan Low.
