In this study, we investigate the genetic variation associated with blocking and use these analyses to generate testable hypotheses surrounding the mechanism of Wolbachia-mediated dengue blocking. From our results, we hypothesize that Wolbachia may block virus replication by increasing the regeneration rate of mosquito cells via the Notch signaling pathway. We also propose that Wolbachia modulates the host’s transcriptional pausing pathway either to prime the host’s anti-viral response or to directly inhibit viral replication.

From 2013 to 2015, sanitary authorities reported an increased incidence of Guillain-Barré syndrome (GBS) associated with Zika virus (ZIKV) in French Polynesia, Caribbean, and Brazil.1–3 After the end of ZIKV epidemics, GBS cases where still above the usual limits in countries where the arrival of chikungunya virus (CHIKV) was also a concern.3
In conclusion, our data provides evidence that CHIKV infection activates the TLR pathways in primary monocytes and MDMs, which play a crucial role in CHIKV pathogenesis and/or host defense, differentially.
