Final Results of a Phase 1 Study of Vorinostat, Pegylated Liposomal Doxorubicin, and Bortezomib in Relapsed or Refractory Multiple Myeloma

We evaluated the combination of pegylated liposomal doxorubicin, bortezomib and vorinostat in 32 patients with relapsed/refractory multiple myeloma in a phase I study. The maximum tolerated dose of vorinostat with PLD/bortezomib was 400 mg on days 4 to 11. Clinical activity was seen including in bortezomib-refractory patients. However, hematologic, constitutional and gastrointestinal toxicity was common.
