We describe here that pre-exposure with a live virus generated via a A/WSN/1933(H1N1) reverse genetics system resulted in a significant reduction of viral shedding from pigs exposed to either a swine H1N1 virus or a swine H3N2 virus. At 3-day post challenge (DPC), approximately 1 log and 1.5 logs reductions of viral shedding were observed in the swine H1N1- and H3N2-challenged vaccinated pigs when compared to unvaccinated animals. This article aims to evaluate a three-stage PEG-Salt aqueous two-phase extraction (ATPE) system to purify foot-and-mouth disease virus (FMDV). Unique separation efficiencies were obtained by selecting phase-forming salt and varying the molecular weight (MW) of PEG and the pH value by using neutral salt additives. Improving the purity of FMDV and removing the phase-forming polymer were performed by multi-stage ATPE system. Results revealed that the partition coefficient (K) of FMDV in single-stage ATPE did not linearly increase with the MW of PEG. CONCLUSIONS: PHDs on remote islands highly value the validity and deployment needs of PHDs, while PHDs in close proximity to private clinics or hospitals give a low score. This suggests that there is a need to more efficiently deploy PHDs depending on local characteristics and the presence or absence of nearby private medical clinics and hospitals.
