{alpha}-Conotoxin Vc1.1 inhibits human dorsal root ganglion neuroexcitability and mouse colonic nociception via GABAB receptors

Conclusions Vc1.1-mediated activation of GABABR is a novel mechanism for reducing the excitability of human DRG neurons. Vc1.1-induced activation of GABABR on the peripheral endings of colonic afferents reduces nociceptive signalling. The enhanced antinociceptive actions of Vc1.1 during CVH suggest it is a novel candidate for the treatment for CVP.

We present the case of a 48-year old male patient with chronic motor complete SCI, who benefited from a 5-day period of bilateral L2-level DRG-stimulation by experiencing suppression of transfer-evoked spasticity problems and of chronic lower back pain. To the best of our knowledge, this is the first case describing the successful application of DRG-stimulation for spasticity depression in patients with chronic SCI.

CONCLUSIONS: SRS did not alter sensory or motor function but reduced the activation of satellite glial cells, a pathway for DRG-mediated pain perpetuation. Radiosurgery provoked changes equivalent to the effects of focal radiation on the trigeminal ganglion after SRS for trigeminal neuralgia, suggesting that radiosurgery could be successful in relieving radiculopathic pain.
