Association of ORAI1 gene polymorphisms with chronic spontaneous urticaria and the efficacy of the nonsedating H1 antihistamine desloratadine

Chronic spontaneous urticaria (CSU) is a common skin disease associated with mast cell activation and degranulation.1,2 The degranulation process of mast cells is mediated mainly by calcium-release –activated calcium channels–mediated Ca2+ influx. ORAI1, encoded by ORAI1 gene, is the pore-forming subunit of calcium-release–activated calcium channels.3-5 A study on ORAI1 knockout mouse demonstrated that ORAI1 is essential for mast cell degranulation, leukotriene (LTC4) secretion, histami ne release as well as TNF-α secretion.
