Ascorbic acid ameliorates behavioural deficits and neuropathological alterations in rat model of Alzheimer ’s disease

Here we investigated the efficacy of ascorbic acid (AA) in reversing aluminium chloride (AlCl3)-induced behavioural deficits and neurotoxic cascades within prefrontal cortex (PFC) and hippocampus of rats. A group of rats administered oral AlCl3 (100mg/kg) daily for 15days showed degenerative changes characterised by significant weight loss, reduced exploratory/working memory, frontal-dependent motor deficits, cognitive decline, memory dysfunction and anxiety during behavioural assessments compared to control. Subsequent analysis showed that oxidative impairment-indicated by depleted superoxide dismutase and lipid peroxidation (related to glutathione-S-transferase activity), cholinergic deficits seen by increased neural acetylcholinesterase (AChE) expression and elevated lactate dehydrogenase underlie behavioural alterations. In this study, 2-month-old triple transgenic AD model mice (3 × Tg-AD) was treated with 0.2 and 1.0 mM bis(ethylmaltolato) oxidovanadium IV (BEOV) in drinking water for 2 months respectively. The results of open field test, elevated plus-maze test and Y maze test showed that BEOV could significantly improve the exploratory ability, memory capacity and exercise ability and relieve the anxiety of 4-month-old 3 × Tg-AD mice. Conclusion: The once-a-week CST approach seems to be a viable alternative to twice-a-week CST for community-dwelling older adults with mild to moderate cognitive deficits. Further investigation is required to determine the optimal dose of CST for the elderly.

Fragile X syndrome (FXS) is the most common inherited form of intellectual disability and is associated with increased risk for ASD, anxiety, ADHD, and epilepsy. Although our understanding of FXS pathophysiology has improved, a lack of validated blood-based biomarkers of disease continues to impede bench-to-bedside efforts. As a biomarker target in FXS, amyloid- β (Aβ) precursor protein (APP) is best understood in the context of Alzheimer disease; there is a growing body of evidence suggesting that the molecule and its derivatives play a broader role in neuronal hyperexcitability, a characteristic of FXS.
