Conclusion: We find that re-expression of silenced MAN1C1 in ccRCC cell lines inhibited cell viability, colony formation, induced apoptosis, suppressed cell invasion and migration. In conclusion, MAN1C1 is a novel functional tumor suppressor in renal carcinogenesis. This is the first time that the function of MAN1C1 gene has been verified in the renal tumor tissue so far.

CONCLUSION: ccRCC cells are characterized by high free iron levels and a cancer-specific dependency on iron for HIF-α overexpression, cell cycle progression and apoptotic escape. This iron dependency is introduced by VHL inactivation, revealing a novel interplay between VHL/HIF-α dysregulation and ccRCC iron metabolism. Future study is warranted to determine if iron deprivation using chelator drugs provides an effective therapeutic strategy for targeting HIF-2α and suppressing tumor progression in ccRCC patients. Kidney cancer is among the 10 most common human malignancies in men and women [1,2]. According to statistics, approximately 65,340 new cases and 14,970 deaths are related to this malignancy in 2018 in the United States [3]. Renal cell carcinoma (RCC), which is derived from renal tubular epithelial cells, is the most common histological type and accounts for over 80% of all cases [1,2,4,5]. Approximately more than 15% of all RCC patients are diagnosed with metastatic renal cell carcinoma (mRCC) [1,3,6], and over 20% of patients with localized RCCs have metastases after partial or radical nephrectomy [7].
